State Route 316 (SR 316) is a 14.05-mile east–west highway in Pickaway County, Ohio. It is maintained by the Ohio Department of Transportation (ODOT). The road begins at SR 56 in the Five Points area of Monroe Township and ends at SR 752 in Ashville, passing through Darbyville and South Bloomfield along the way.

From Five Points, SR 316 travels east-northeast through farmland for about 4 miles, enters Darbyville, crosses the Big Darby Creek, then continues east, crossing the Scioto River into South Bloomfield. In South Bloomfield, it meets US 23 (Walnut Street) at a traffic signal, briefly runs south with US 23, then turns east onto Ashville Road. The route becomes Pickaway Street, enters Ashville as West Main Street, and in the town turns north on Long Street to end at SR 752; beyond the end, the road continues as County Route 28.

SR 316 began as a 1.5-mile spur from US 23 in South Bloomfield to Ashville in 1931. By 1937, it was extended west to Five Points at SR 56. The connection between downtown Ashville and SR 752 was added around 1970. The entire route is in Pickaway County.
