Ohio State Route 159 (SR 159) is a roughly 31-mile state highway in south-central Ohio, running northeast to southwest. It is maintained by the Ohio Department of Transportation (ODOT). The southern end is in Chillicothe at the US 50 and SR 104 interchange, and the northern end is at US 22 about 4 miles southwest of Lancaster. SR 159 passes through Ross, Pickaway, and Fairfield counties.

Key connections include US 35 in Chillicothe, US 23 near Chillicothe, SR 207 near Chillicothe, and SR 180 near Kingston.

History in brief:
- Signed in 1923 along most of its current route.
- Extended south to US 23 in 1927 (along the former SR 27).
- The first paved section appeared in 1933 (southern end to Meade); then Meade to Tarlton in 1935; Tarlton to the northern end in 1938.
- Extended north to the current terminus along the former US 22 in 1971; extended south to the current terminus along the former US 23 in 1972.
- A Ross County project added two roundabouts at the new intersection with SR 207 and at the SR 180 intersection, built from Oct 2019 to Oct 2020.
