Kei Nishikori defended his Memphis Open singles title in 2016, beating Taylor Fritz 6–4, 6–4 in the final. This was Nishikori’s third straight Memphis Open title. Fritz was making only his third career ATP Tour appearance. The tournament drew 28 players in singles, with 8 seeds; the top four seeds received byes into the second round.
