Giovanni Vincenzo Acquaviva d'Aragona was a cardinal of the Roman Catholic Church who lived in Italy. He was born in Naples sometime between 1490 and 1495 and died on 16 August 1546 in Itri. He came from a powerful southern noble family; his father was Andrea Matteo III Acquaviva d'Aragona, the 8th Duke of Atri, and his mother was Isabella Piccolomini. His brother Giannantonio Donato’s son, Claudio Acquaviva, became the fifth General of the Jesuits, making Claudio Giovanni’s nephew.

In February 1537, Giovanni was elected bishop of Melfi and Rapolla. In June of that year, he became Archpriest of Santa Maria in Platea di San Flaviano. Pope Paul III raised him to the rank of cardinal on 2 June 1542 and appointed him Prefect of Castle Sant'Angelo; ten days later he received the titular seat of San Martino ai Monti. Giovanni Vincenzo Acquaviva d'Aragona died in Itri on 16 August 1546. He was the grand-uncle of later cardinals Giulio Acquaviva d'Aragona and Ottavio Acquaviva d'Aragona.
