Cheyenne Wells is a small statutory town and the county seat of Cheyenne County, Colorado, United States. The population was 758 at the 2020 census. The town covers about 1.07 square miles (2.77 square kilometers), all land, and sits at an elevation of 4,292 feet.

The town’s name comes from wells once used by the Cheyenne people. The Cheyenne post office opened in 1876. Cheyenne County was created in 1889 with Cheyenne Wells as the county seat, and the town was incorporated on May 14, 1890.

Geographically, Cheyenne Wells lies at 38.8214°N, 102.3532°W. A small area about 10 miles southwest of town is opposite Île Saint-Paul in the southern Indian Ocean.

U.S. Highway 40 runs through the community, and the Union Pacific Railroad passes nearby. The Ladder Creek Helium Plant near Cheyenne Wells is owned by Tumbleweed Midstream and is one of only about fourteen helium plants in the world.
