State Route 161 (SR 161) is an east–west highway in central Ohio. It runs about 58 miles from Mutual at SR 29 in the west to near Alexandria at SR 37 in the east. The route passes through northern Columbus and towns such as Plain City, Dublin, and New Albany.

SR 161 was created in 1924 to connect Plain City and Dublin and was later extended west to Mutual and east to Granville. In Dublin and New Albany, parts of the route were upgraded to freeways (1969 in Dublin and 1997 around New Albany). A bypass around New Albany opened in 2000. From 2004 to 2008, the Northeast Expressway Transformation rebuilt many bridges and ramps, and the freeway portion was extended to SR 37.

The highway is part of the Big Darby Plains Scenic Byway on some segments. An unsigned SR 161J existed in Licking County from 2008 until it was abandoned in 2013.
