Cobleskill Creek is a river in central New York that runs through Otsego and Schoharie counties. It begins on Pine Mountain, west of West Richmondville, and flows east-northeast until it joins Schoharie Creek in Central Bridge. From there, the water continues through the Mohawk and Hudson rivers toward Upper New York Bay. The creek passes through the towns of Richmondville and Cobleskill, and its drainage basin covers about 139 square miles.

Its main tributaries are West Creek from the left and Punch Kill and Bear Gulch Brook from the right. The water from Cobleskill Creek eventually reaches the Hudson River and New York Bay.

The U.S. Geological Survey tracks stream flow along the creek. A gauge on South Grand Street, about 0.4 miles south of Cobleskill, has operated at various times since 1963. It recorded a maximum flow of about 6,570 cubic feet per second in March 1964 and a minimum flow of about 2.1 cubic feet per second in July 2018.
