Sainte-Gemme-Martaillac is a small commune in south-western France, in the Lot-et-Garonne department of the Nouvelle-Aquitaine region. It covers 114 square kilometers and had 377 residents in 2023, making it sparsely populated. The landscape ranges from 58 to 156 meters above sea level (about 80 meters on average).

The commune is part of the Marmande arrondissement and the canton of Les Forêts de Gascogne, within the Coteaux et Landes de Gascogne intercommunality.
